Comparison of Polymorphisms in Genes Encoding Serum Matrix Metalloproteinase-1 and -12 between Patients with Invasive Pituitary Macroadenomas and Healthy Individuals

Abstract
Aim
Pituitary adenomas may present invasive features, accounting for a considerable number of intracranial malignancies. Matrix metalloproteinases (MMPs) are endopeptidases that play key roles in extracellular matrix remodeling, tumor invasion, and metastases. Genetic polymorphisms in MMP genes might influence tumor behavior. This study investigated the association between invasiveness of nonfunctional invasive pituitary macroadenomas and gene polymorphisms in MMP1 and MMP12.
Material and Methods
The overall cohort comprised 50 healthy individuals and 75 patients diagnosed with invasive nonfunctional pituitary macroadenomas. DNA was extracted from blood samples, the specific polymorphic regions were amplified by PCR, and genotyping was performed using restriction fragment length polymorphism analysis. Gene polymorphism distributions were compared using the chi-square test.
Results
The patient and control groups showed no significant variation in the genotype distribution of MMP1 (−519) and MMP12 (−82 A>G) polymorphisms (p = 0.831 and p = 0.541, respectively). Moreover, the genotype distributions of MMP1 and MMP12 polymorphisms were independent of age and sex.
Conclusion
No significant association was found between invasive nonfunctional pituitary macroadenomas and gene polymorphisms in MMP1 and MMP12. Future studies should incorporate larger cohorts and patients with different tumor grades to clarify the genetic mechanisms influencing the invasiveness of pituitary adenomas.
